Restore wallet/coins from back-up

Hi all, bit of a newbie question.

Situation as follows:

• I had bitcoin in my Bitcoin-QT wallet which i then transferred to another address
  The coins never appeared at the destination address
  Sure enough, the transaction doesn't appear on blockchain.info so i understand it has not been recognized by the blockchain, but my balance is 0.
  I have a backup copy of my wallet.dat file from a time prior to this failed transaction.

To recover my BTC, so i just need to copy the back up file into the current wallet location and sync?
Can I use the back up in a completely new location (i.e. a new instance of a bitcoin client on another machine) and just restore my balance there?

Just trying to work out best way to recover my BTC and don't want to do the wrong thing and end up losing them!
